70% of the new synthesized small molecules are poorly soluble compounds, resulting in a low bioavailability. Bioavailability enhancement is the number one challenge in API formulation development today.
One of the main approaches to increase the solubility is by creating an amorphous solid dispersion, which can be created by different technologies such as spray-drying, spray-congealing, fluid bed bead coating, Hot melt extrusion. After processing, the API is dispersed on molecular level in a polymeric matrix. The polymer is used to stabilize the amorphous, metastable form of the drug and to sustain supersaturation of the API in biological fluids, thereby increasing the bioavailability.
